Sulfur-containing Proteins and Epidermal Keratinization

چکیده
Sites of synthesis of sulfur-containing protein in keratinizing epithelium have been studied by radioautography after injection of sulfur-labeled cystine. Bern et al. (3) demonstrated concentration of silver grains immediately below the cornified layer, suggesting that much of sulfur is added at the final stages of keratinization as observed in hair. In contrast, B6langer (4) reported that protein synthesis occurs mainly in the Malpighian cells, and Hoober and Bernstein (17) reached a similar conclusion. Radioautographs prepared with beta emission of sulfur 35 lack resolution, and determination of precise localization of radioactivity with sulfur 5 becomes very difficult because of its greater energy (167 Kev) compared to that of tritium (18 Kev) (24). In addition, light microscopy alone is not sufficient to study distribution and movement of newly synthesized protein,

High TMEM45A expression is correlated to epidermal keratinization.
TMEM45A (DERP7, DNAPTP4 or FLJ10134) gene, belonging to the TMEM family encoding predicted transmembrane proteins, is highly expressed in epidermal keratinocytes. To investigate the potential involvement of TMEM45A during the differentiation and keratinization processes, its expression has been characterized in normal human keratinocytes and the protein subcellular localization has been studied...
متن کاملInvited review: formation of keratins in the bovine claw: roles of hormones, minerals, and vitamins in functional claw integrity.
Keratins are the characteristic structural proteins of the highly cornified epidermis of the skin, feathers, and hoof. Keratin proteins provide the structural basis for the unique properties of the biomaterial horn and its protective function against a wide range of environmental factors. Hoof horn is produced through a complex process of differentiation (keratinization) of epidermal cells. For...
متن کاملHIGH-PERFORMANCE LIQUID CHROMATOGRAPHIC ANALYSIS OF SULFUR MUSTARD REACTION WITH AMINO ACIDS AND PROTEINS
The interaction of sulfur mustard with aminoacids and proteins has been investigated in this study. Rats were injected with sublethal doses of sulfur mustard subcutaneously and intraperitoneally. At different time intervals, plasma and urine samples were collected. The binding affinity of sulfur mustard with urinary and plasma proteins and enzymes was studied for the first time using non-i...
متن کاملTMEM45A Is Dispensable for Epidermal Morphogenesis, Keratinization and Barrier Formation
TMEM45A gene encodes an initially uncharacterized predicted transmembrane protein. We previously showed that this gene is highly expressed in keratinocytes where its expression correlates with keratinization, suggesting a role in normal epidermal physiology. To test this hypothesis, we generated TMEM45A knockout mice and found that these mice develop without any evident phenotype. The morpholog...
متن کاملSpecific alterations in keratin biosynthesis in mouse epidermis in vivo and in explant culture following a single exposure to the tumor promoter 12-O-tetradecanoylphorbol-13-acetate.
Treatment of mouse skin with a single dose of the phorbol ester tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA) causes dramatic alterations in the biosynthesis of specific epidermal keratins. TPA was applied either topically to the skin of mice or added to the culture medium of skin explants maintained in vitro. Twenty-four h after exposure, skin samples were pulse labeled with [35S]m...
